Anti-H3-K4-HMTase SETD7抗體,組蛋白H4-K4甲基轉移酶抗體一抗孵育(Primary antibody incubation):
參考一抗的說明書，按照適當比例用Western一抗稀釋液稀釋一抗。
用微型臺式真空泵吸盡封閉液，立即加入稀釋好的一抗，室溫或4℃在側擺搖床上緩慢搖動孵育一小時。如果一抗孵育一小時效果不佳，可以在4℃緩慢搖動孵育過夜?；厥找豢?。加入Western洗滌液，在側擺搖床上緩慢搖動洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結果背景較高可以適當延長洗滌時間并增加洗滌次數。

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

英文介紹：Histone methyltransferase that specifically monomethylates 'Lys-4' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. Plays a central role in the transcriptional activation of genes such as collagenase or insulin. Recruited by IPF1/PDX-1 to the insulin promoter, leading to activate transcription. Has also methyltransferase activity toward non-histone proteins such as p53/TP53, TAF10, and possibly TAF7 by recognizing and binding the [KR]-[STA]-K in substrate proteins. Monomethylates 'Lys-189' of TAF10, leading to increase the affinity of TAF10 for RNA polymerase II. Monomethylates 'Lys-372' of p53/TP53, stabilizing p53/TP53 and increasing p53/TP53-mediated transcriptional activation. Also able to demethylated 'Lys-372' of p53/TP53 in vitro.
